Theory of operating HPLC column in reversed flow direction
Many operators try to reverse the flow direction of HPLC column to obtain acceptable system suitability results such as ( Tailing factor, Plate count and resolution ) - In your opinion, Do you think that this actually affects on system suitability ? - What is the scientific theory of this ? - Do you think that this will affect on the column performance on the long run 🤔? " Lets discuss this with each other in the comments 👇"

I think front tailing is usually due to the solvent effect try either to minimize the injection volume or to change the last dilution of test to be in mobile phase and not in diluent
ReplyDeleteYes I totally agree with you, also it may be because the column loses its efficiency
